A DNA fragment of unknown sequence is divided into four portions and mixed with all the elements necessary to synthesize the complementary strand according to the Sanger sequencing method. To each of these four mixtures a different dideoxynucleotide (a modified base) is added in addition to the normal nucleotides.  Each mixture is then separated by electrophoresis and the gel shown below is obtained.
