Neurоfibrоmаtоsis type 1 (NF1) is а tumor predisposition syndrome thаt is inherited in an autosomal dominant pattern. NF1 is characterized by the development of neurofibromas, which are benign tumors that develop on nerves, as well as other clinical criteria (e.g., the presence of café-au-lait macules (CALMs), which are flat, discolored birthmarks that are usually light brown in color). Mutations in the NF1 gene cause NF1. More than 3000 different genetic mutations in the NF1 gene have been reported. In 2022, a study examined genotype-phenotype correlations in NFT1. Consider the data shown below which has been taken from that 2022 study: Patient 1, 2 and 3 all have the same mutation in NF1 - a 'C' at position 3826 in the DNA sequence is changed to a 'T'. However, while these patients have the exact same mutation, they don't show the exact same clinical features. For example, patient 1 and 2 have cardiovascular disorders, and patient 3 does not. Patient 3 has pheochromocytoma/paraganglioma (tumor in the adrenal glands/neuroendocrine tumor) but patient 1 and 2 do not.
