To achieve this, they immersed the chloroplasts in a pH 4 solution for about 10-15 seconds. The low pH solution was able to cross the membrane, lowering the pH inside the thylakoid lumen. Next, the scientists added ADP and inorganic phosphate (Pi) to the solution and then rapidly increased the pH to 8, creating a pH gradient across the membrane.
